ABSTRACT

Purpose: To evaluate whether the toric intra?ocular lens (IOL) power calculation based on total corneal astigmatism (TCA) in eyes with high posterior corneal astigmatism (PCA) could result in a systematic over?correction or under?correction after operation. Methods: The present study included a mono?centric retrospective study design. The data were collected from 62 consecutive eyes during uncomplicated cataract surgery by a single surgeon with a measured PCA of 0.50 diopters (D) or higher. Toric IOL calculations were made using TCA measurements. The eyes were grouped as either “with?the?rule” (WTR) or “against?the?rule” (ATR) on the basis of the steep anterior corneal meridian. The post?operative refractive astigmatic prediction error was analyzed 1 month post?operatively using the vector analysis by the Alpins method and double?angle plots method. Results: The correction indexes were 1.14 ± 0.29 in the ATR eyes and 1.25 ± 0.18 for the WTR eyes, indicating a tendency toward over?correction. The mean over?correction was 0.22 ± 0.52D in the ATR group and 0.65 ± 0.60D in the WTR group. The magnitude of error (ME) values were significantly different from the ideal value of zero in both groups (ATR: P = 0.03; WTR: P = 0.00). No significant difference in mean absolute error (MAE) in predicted residual astigmatism was found between ATR and WTR groups (0.61 ± 0.42 D versus 0.64 ± 0.39 D; P = 0.54). The ATR group yielded better results, with 48% <0.50D prediction error in the main analysis. Conclusions: The results suggested that in cases of high PCA, the toric IOL calculation, which was performed using TCA, may cause a potential over?correction in the ATR and WTR eyes. For ATR eyes, over?correction led to slight disruption of post?operative visual quality because of the “with?the?rule” residual astigmatism after operation. Therefore, we suggested using TCA for toric IOL calculation in ATR eyes.

ABSTRACT

Purpose: To evaluate a method using measured values of total corneal refractive power (TCRP) for a manufacturer’s online calculator by comparing it with the Barrett toric calculator (BTC) and Kane toric calculator (KTC) combined with simulated keratometry values (SimK). Methods: This was a retrospective case series. Patient records were reviewed to identify the patients who had biometry with the IOL Master 700 and Pentacam recorded before toric IOL implantation and refractive follow?up data after implantation. The predicted error in residual astigmatism was calculated by vector analysis according to the calculation methods and the measurements used. Results: A total of 70 eyes of 56 patients were included. The mean absolute astigmatism prediction errors were 0.6 ± 0.32, 0.59 ± 0.35, and 0.61 ± 0.35 D for the ATCTCRP, BTCSimK, and KTCSimK calculators, respectively (P = 0.934), and the centroid of the prediction errors were 0.3 D @ 178°, 0.11 D @ 102°, and 0.09 D @ 147°, respectively (P = 0.23). In the with?the?rule subgroup, the centroid of the prediction error was 0.34 D @ 176° for ATCTCRP and was the highest among the three calculation methods (P = 0.046). Conclusion: The ATCTCRP, BTCSimK, and KTCSimK calculators had similar performance with regards to their astigmatism prediction accuracy. The ATCTCRP calculator combined with 4.0?mm apex/ ring readings of TCRP was slightly intended to result in against?the?rule residual astigmatism.

@#以程序性死亡因子 1(PD-1)及其配体 1(PD-L1)为代表的B7/CD28家族在肿瘤免疫治疗中显示出极大的应用潜力,但仍 有很多PD-L1检测阴性患者无法从中受益。作为B7家族成员,人内源性逆转录病毒-H长末端重复关联蛋白2 (HHLA2)分子结 构与PD-L1等其他B7家族成员有一定的同源性和相似性, 对T细胞有共刺激和共抑制作用,在多数实体肿瘤中呈高表达,且在某 些实体肿瘤中表达比PD-L1更广泛;其通过与受体穿膜和免疫球蛋白结构域2(TMIGD2)结合可促进肿瘤免疫逃逸,导致肿瘤的 进展和转移。基于此,HHLA2/TMIGD2有望成为肿瘤免疫治疗新的通路和靶点之一。本文就HHLA2及其受体TMIGD2的分子 结构、生物学功能及其在实体肿瘤中作用的研究进展作一综述。

@#近年来 DNA修复途径成为癌症治疗的热门靶点, 聚ADP核糖聚合酶[poly(ADP)-ribosepolymerase,PARP]作为DNA修 复的关键酶得到了广泛研究,目前上市的PARP抑制剂(PARP inhibitor,PARPi)药物,在乳腺及卵巢等肿瘤中取得了革命性的突 破。结直肠癌(CRC)具有异质性,其发生与发展是一个多途径、多基因参与及多步骤的过程,其发病率与病死率在中国逐年上 升。研究发现,PARP1与CRC的发生、发展及治疗密切相关,本文从PARP1的分子结构及生物学功能、PARPi的作用机制、PARP1 在CRC组织中的表达情况、PARP1与结直肠癌干细胞的关系、PARPi与CRC治疗的关系等5个方面对PARPi在CRC中作用的研 究进展作一系统综述,为临床治疗CRC寻找新的治疗策略。

ABSTRACT

Objective@#The aim of the study was to identify the personality types of middle school students and its association with life satisfaction,and to provide a reference for improving personality and mental health of middle school students.@*Methods@#A total of 1 409 students randomly selected form 4 middle schools of a rural area in Wuhan city by cluster stratified sampling method were taken as the object of the research. Adolescents completed the Chinese version of NEO Five-Factor Inventory (NEO-FFI) and the Satisfaction with Life Scale. Latent profile analysis (LPA) was used to identify personality types. ANOVA analyses were used to explore the associations between personality types and life satisfaction.@*Results@#Three profiles were identified: resilient (5.32%), overcontrolled (32.86%) and undercontrolled (61.82%),the differences were of statistical significance(P<0.05). Resilient adolescents had the highest scores (M=5.10, SD=1.34) whereas undercontrolled had the lowest scores (M=3.66, SD=1.09) on life satisfaction.@*Conclusion@#Schools, families and society should pay attention to the high proportion of overcontrolled and undercontrolled in middle school students and take actions to promote the positive development of personality and mental health.

ABSTRACT

BACKGROUND: Drought is a major abiotic stress affecting global wheat (Triticum aestivum L.) production. Exploration of drought-tolerant genes is essential for the genetic improvement of drought tolerance in wheat. Previous studies have shown that some histone encoding genes are involved in plant drought tolerance. However, whether the H2B family genes are involved in drought stress response remains unclear. METHODS: Here, we identified a wheat histone H2B family gene, TaH2B-7D, which was significantly up-regulated under drought stress conditions. Virus-induced gene silencing (VIGS) technology was used to further verify the function of TaH2B-7D in wheat drought tolerance. The phenotypic and physiological changes were examined in the TaH2B-7D knock-down plants. RESULTS: In the TaH2B-7D knock-down plants, relative electrolyte leakage rate and malonaldehyde (MDA) content significantly increased, while relative water content (RWC) and proline content significantly decreased compared with those in the non-knocked-down plants under drought stress conditions. TaH2B-7D knock-down plants exhibited severe sagging, wilting and dwarf phenotypes under drought stress conditions, but not in the non-knocked-down plants, suggesting that the former were more sensitive to drought stress. CONCLUSION: These results indicate that TaH2B-7D potentially plays a vital role in conferring drought tolerance in wheat.

ABSTRACT Antiviral drug resistance is the most important factor contributing to treatment failure using nucleos(t)ide analogs such as lamivudine for chronic infection with hepatitis B virus (HBV). Development of a system supporting efficient replication of clinically resistant HBV strains is imperative, and new antiviral drugs are needed urgently to prevent selection of drug-resistant HBV mutants. A novel fluorinated cytidine analog, NCC (N-cyclopropyl-4′-azido-2′-deoxy-2′-fluoro-β-d-cytidine), was recently shown to strongly inhibit human HBV in vitro and in vivo. This study was designed to evaluate the antiviral activity of NCC against lamivudine-resistant HBV. We generated a stable cell line encoding the major pattern of lamivudine-resistant mutations rtL180M/M204V and designated it "HepG2.RL1". Immuno-transmission electron microscopic examination and enzyme-linked immunosorbent assay were used to detect secretion of HBV-specific particles and antigens. Quantification of extracellular DNA and intracellular DNA of HepG2.RL1 cells by quantitative real-time polymerase chain reaction revealed >625-fold and >5556-fold increases in the 50% inhibitory concentration of lamivudine, respectively, compared with that for the wild-type virus. The results showed that NCC inhibited DNA replication and HBeAg production in wild-type or lamivudine-resistant HBV in a dose-dependent manner. In conclusion, screening for antiviral compounds active against lamivudine-resistant HBV can be carried out with relative ease using hepG2.RL1 cells. NCC is a potential antiviral agent against wild-type HBV and clinical lamivudine-resistant HBV and deserves evaluation for the treatment of HBV infection.

Objective @# To investigate the effects of different root canal filling stop on quality of root canal filling and apical sealing in root canals obturated with GuttaFlow. @* Methods@#60 teeth were randomly divided into three groups, using different root canal filling stops to shape the root canals with MTwo (25/06) file. All root canals were obturated with Gutta Flow, and the overfilling of the root canals were recorded and evaluated by X-ray. And the apical microleakage of teeth was evaluated by transparent teeth technique.@*Results@#The roots were prepared with MTwo (25/06) as master apical file, the overfilling rate of the root canals in root canal filling stop was higher as the distance from the apex was shorter, but there was no significant difference. The under-filling rate of the root canals in root canal filling stop was higher as the distance from the apex was longer. And the under-filled root canals in root canal filling stop 0.5 mm from the apex showed a statistically significant difference with 2 mm. The mean dyeing penetration length in 0.5 mm and 1 mm group was significantly shorter than 2 mm group. @*Conclusion @#A suitable root canal filling stop could improve the quality of root canal filling in root canals obturated with GuttaFlow.

ABSTRACT

Since the diccovery of neural stem cells (NSCs) in the embryonic and adult mammalian central nerous system, it provided novel ideas forneurogenesis as the potential of proliferation and differentiation of NSCs. One of the ways to promote the clinical application of neural stem cells (NSCs) is searching effective methods which regulate the proliferation and differentiation.This is also a problem urgently to be solved in medical field. Plenty of earlier studies have shown that traditional chinese medicine can promote the proliferation and differentiation of NSCs by regulating the related signaling pathway in vivo and in vitro. The reports of Chinese and foreign literatures on regulating the proliferation and differentiation of neural stem cells in recent ten years and their target and signaling pathways is analyzed in this review.The traditional chinese medicine regulate proliferation and differentiation of NSCs by the signaling pathways of Notch,PI3K/Akt, Wnt/β- catenin, and GFs. And, those signaling pathways have cross- talk in the regulation progress. Moreover, some traditional Chinese medicine, such as astragalus,has a variety of active ingredients to regulate proliferation and differentiation of NSCs through different signaling pathways. However, to accelerate the clinical application of neural stem cells, the studies aboutthe proliferation and differentiation of NSCs and Chinese medicine should be further deepened, the mechanism of multiple targets and the comprehensive regulation function of traditional Chinese medicine should be clarified.

Background: Paracoccidioidomycosis (PCM) is a neglected systemic mycosis caused by a dimorphic fungus of the Paracoccidioides genus. The standard diagnosis is based on isolation of the fungi in culture, and by microscopic visualization of characteristic multiple budding yeast cells in biological samples. However, in some situations, access to the site of injury prevents the collection of biological material. A variety of immuno-serological techniques has proven useful for allowing inferring diagnosis with a certain degree of certainty, thus optimizing time. The aim of this study was to standardize and validate the Dot-ELISA (DE) assay, comparing it with the serological standard, double immunodiffusion (DI). Methods: In order to standardize the DE assay, 143 serum samples were used. Out of those, 23 were from apparently healthy patients, 77 were from patients with confirmed PCM and 43 were from patients with other lung infections (tuberculosis, aspergillosis and histoplasmosis). To validate the DE technique, 300 serum samples from patients with PCM clinical suspicion (probable and possible cases) were employed, and these results were compared with those of DI. Results: The DE assay showed sensitivity of 91%, specificity of 95.4%, positive predictive value of 96%, negative predictive value of 98.2%, accuracy of 93%, and great precision (k = 0.93). In addition, the nitrocellulose membranes have proved to be viable for using at least 90 days after P. brasiliensis B-339 antigen sensitization. Conclusion: Dot-ELISA method was found to be an extremely promising tool as serologic screening technique, because of its high sensitivity. Furthermore, Dot-ELISA shows the prospect of being transferred to laboratories of mycoserology including those with fewer resources or even to be used directly in the field. It has an excellent shelf life ­ membranes coated with antigen can be used for testing without changes in the pattern of reactivity among laboratories ­ and presents reliable values of sensitivity, specificity, predictive values, accuracy and a high correlation with the serological standard methodology. Based on the present findings, it possible to state that this technique constitutes a remarkable option to be used in routine diagnosis for public health centers.(AU)

Abstract Background: The gamma-type phospholipase A2 inhibitor (LI) is a natural protein commonly found in snake serum, which can neutralize pathophysiological effects of snake venom phospholipases A2. Therefore, this protein is a potential candidate to the development of a novel antivenom. To the best of our knowledge, there is no antibody currently available for PLI identification and characterization. Methods: Bioinformatics prediction of epitope using DNAStar software was performed based on the sequence of Sinonatrix annularis PLI (SaPLI). The best epitope 151CPVLRLSNRTHEANRNDLIKVA172 was chosen and synthesized, and then conjugated to keyhole limpet hemocyanin and bovine serum albumin for use as an immunogen and plate-coating antigen, respectively. Results: Eighteen IgG anti-PLI mAb hybridoma cell strains were obtained, and all the mAbs had positive interaction with recombinant His6-PLI and natural SaPLI. Moreover, the mAb from 10E9 strain was also successfully used for the immunodetection of other snake serum PLIs. cDNA sequence alignment of those PLIs from different snake species showed that their epitope segments were highly homologous. Conclusions: The successful preparation of anti-PLImAb is significant for further investigation on the relationship between the structure and function of PLIs, as well as the interaction between PLIs and PLA2s.

Objective To analyze the characteristics of clinical manifestations, risk factors, therapies and acute outcomes in patients with cerebral venous sinus thrombosis complicated by cerebral hemorrhage. Methods Seventy-five patients with cerebral venous sinus thrombosis were included in the study. According to the radiological findings on the brain image, patients were divided into two subgroups:cerebral hemorrhage group and non-hemorrhage group. The demo?graphic data, potential risk factors, clinical manifestations, radiological features, therapeutic strategies and acute out?comes were compared between two subgroups, and high risk factors were also analyzed. Results There were seventy-five patients with cerebral venous sinus thrombosis in the present study. Twenty-eight patients of them (37.2%) had cerebral hemorrhage whereas the remaining forty-seven patients (62.7%) did not have cerebral hemorrhage. Pregnancy/puerperi?um were significantly higher in patients with cerebral hemorrhage (with vs without;28.6%vs. 6.4%, P=0.015), while in?fection was markedly higher in patients without cerebral hemorrhage (with vs without;7.1% vs. 29.8%, P=0.021). Head?ache (92.9% vs. 70.2%, P=0.021), unconsciousness (25.0% vs. 6.4%,P=0.034), seizures (53.6% vs. 19.1%, P=0.002) and motor deficits (35.7% vs. 12.8%, P=0.019) were more common in patients with cerebral hemorrhage. Moreover, mul?tiple sinus involvement (1.4% vs. 44.7%, P=0.024) was significantly higher and the acute outcomes(mRS≥3: 46.4%vs.17.0%, P=0.006)were poorer in patients with cerebral hemorrhage. Binary Logistic analysis showed that pregnancy/pu?erperium (P=0.004) and multiple sinus involvement were positively, whereas infection was negatively correlated with cere?bral venous sinus thrombosis and hemorrhage ( P=0.007;P=0.03). Conclusions Pregnancy/puerperium, headache, uncon?sciousness, seizures, motor deficits and multiple sinus involvement are more frequently in patients with cerebral venous sinus thrombosis and hemorrhage, and the acute outcomes are poorer in patients with cerebral venous sinus thrombosis complicated by cerebral hemorrhage.
